The research focus of Dr. Medina Franco is computer-aided drug design. While working at the Torrey Pines Institute his research group made significant contributions to identify novel inhibitors of the protein kinase B. His group at the Torrey Pines was one of the first research groups working on computational approaches to identify and optimize the activity of inhibitors of the emerging epigenetic targets DNA methyltranasferases (DNMT). To advance this research project Dr. Medina Franco has had extensive collaborations during the past 8 years with research groups in the US, Mexico, Germany, and Italy experts on DNA methylation and epigenetics. He and his experimental colleagues identified the first selective inhibitor of DNMT3b. In addition, he has implemented computational protocols that have identified novel inhibitors of DNMTs. Dr Medina Franco continued his research on DNMT inhibitors at the Mayo Clinic and the National Autonomous University of Mexico (UNAM).During the past 10 years Dr. Medina Franco has been heavily involved in developing and applying chemoinformatic approaches for drug discovery. In particular his research has focused on structure-activity relationships (SAR) studies using the concept of activity landscape modeling and characterization of the chemical diversity and coverage in chemical space of compound collections. In 2009 he and his colleagues introduced the notion of using multiple molecular fingerprint representations in activity landscape models. Since then, the concept of “consensus” activity cliffs has largely influenced the methodology to develop robust activity landscapes. In 2011 Dr. Medina Franco and Dr. Perez Villanueva, developed the Dual-Activity Difference (DAD) maps and related approaches to systematically explore the SAR of data sets with one or two biological endpoints. These methods are increasingly being used and adapted to explore the SAR of chemogenomics data sets.
