ABOUT YANBO ZHANG

Team and Biology Lead specializing in non-clinical research, from early discovery throughout IND approval, for drug development targeting immunology and inflammation (I&I) diseases.Immunologist by training in adaptive immunity (T cell, Treg) and innate immunity (macrophage), at the levels of molecular, cellular, and systems immunology; working on both in vitro/ex vivo human and murine immune cell-based assays and in vivo disease models.* Specialties: immune tolerance, Regulatory T cell biology, myeloid cell biology, immuno-oncology, autoimmune diseases.Highlights:* CRISPR/Cas9 KO for genetic screening (pool and individual), target discovery and validation in primary immune cells in vivo and in vitro/ex vivo, and mechanism of action studies;* Characterization of various immune cell subsets in vivo and in vitro, using multi-color flow cytometry, immunophenotyping, RNAseq, and analysis (differential expression, signature, enrichment, etc.).* Various mouse disease models: tumor, autoimmune disease, infectious disease, colitis, bone marrow chimeras;* Primary immune cell isolation, sorting, and manipulation;* Primary immune cell culture (T cells, Regulatory T cells /Tregs, macrophages, DCs), various immune cell-based functional assays, and cell-based assay development.* Gene overexpression and genetic knock-out in primary immune cells and various cell lines;* Hematopoietic stem cell (HSC) isolation, genetic manipulation, and transplantation;* Working with PBMC and patient blood samples;* End-to-end lentivirus library and NGS sequencing library construction.* Former trainee of Dr. Christophe Benoist and Dr. Diane Mathis at the Department of Immunology, Harvard Medical School.
