I am a protein biochemist and a mechanistic enzymologist, with an in-depth understanding of chemical biology tools applied to drug discovery and early development. I consider myself as an adaptive team player with resourceful skills in tackling scientific problems.Technical Expertise:• Assay development: Continuous assays using UV/Vis- and fluorescence spectroscopy, HPLC-based discontinuous assays, ALPHA screens, Luminescence - based assays, NMR and LC-MS-based assays, and coupled assays. Gel-based assays and ELISA to quantitate cellular proteins. • Enzyme assays and mechanistic studies: Steady-state enzyme kinetics and inhibition experiments including competitive, non-competitive, allosteric, and mechanism-based inhibitors.• Biophysical assays: Assaying Protein-protein interaction using FRET, ITC, and Biolayer interferometry. CD experiments to evaluate the protein structural perturbations. • Protein chemistry: Expression from bacterial and mammalian systems, purification (AKTA and BioRad), analysis and characterization using SDS PAGE and western blot techniques. • Data and statistical analysis: Proficiency in data analysis using JMP, Graph Pad, Grafit, Kintek and Dynafit software. • Molecular modeling of proteins PyMol, Chimera, and Discovery studio platforms. • Molecular Cloning: Experienced in generating constructs using PCR techniques. DNA sequencing analysis and Gene knockout experiments. • Mammalian cell culture: Proficiency in handling multiple cell lines and performing transfection. chemical biology principles involving bio-orthogonal reactions using fluorescence and affinity probes. • Method development: Quantitative profiling of post-translational modifications, transglutaminases, and peptidases using covalent inhibitors/probes from mammalian cells, tissue, serum, and plasma. • Designing chemical probes and inhibitors: Expertise in developing covalent inhibitors with clickable handles tom function as activity-based probes. Extensive experience in characterizing the inhibition profile of non-reversible and irreversible inhibitors. • Ability to translate scientific questions into analytical strategies.• Effective collaborative skills across teams• Excellent written and communication skills.
