Assembled and led a team of scientists to execute three cutting-edge research projects focused on the mammary-local and systemic immune landscapes and immunotherapeutic responses.I. Breast cancer progression: Discovered and extensively characterized a novel subset of regulatory T cells across five clinical cohorts encompassing approximately 2,100 breast cancer patients, with validation in outbred rat tumor models. Engineered and refined four immunotherapeutic combinations that effectively counter Treg-mediated immune escape, boosting the efficacy of existing PDL1/PD-1-based regimens by 140–210%.II. LFS susceptibility: Elucidated the immune histopathology of both tumor and non-tumor mammary tissues in young women diagnosed with the exceedingly rare Li-Fraumeni Syndrome (LFS) and TP53 germline mutations. Developed and functionally characterized a novel animal model for drug testing of an array of T-cell and dendritic cell-based Immune & Pregnancy: Established analytical pipelines for cryopreserved biospecimens from pregnant women and for samples from pregnant rats. Leveraged proteomic profiling for cross-species comparisons of immune dynamics during pregnancy with a dedicated focus on mammary peptidomes and immune cells. Conducted targeted cell-based depletion, pulse-chase, and adoptive transfer studies to delineate the critical roles of mammary T cells during pregnancy.Mentorship & Leadership: Directly mentoring junior scientists, fostering strategic skill transfer and adaptability within a rapidly expanding laboratory environment. Provided comprehensive guidance through technical training sessions, weekly project meetings, and daily supervision to enhance scientific insight, ensure rigorous quality control of experimental protocols, and maintain robust data management practices. Coordinated internal teams and external contractors to streamline cross-institutional collaborations and ensure the operation of outsourcing tasks.
