My current research at PAVIR is understanding how coagulation crosstalk’s with inflammation, wound healing and immunity by defining how thrombin directly cleaves Osteopontin (OPN) to an activated N-terminal domain which exerts new biological functions. Using a mouse strain I designed where OPN is not cleavable by thrombin(OPN-KI), we have shown that in a mouse melanoma model the OPN-KI mice are protected from metastasis and tumour burden. Several disease indications have been assessed using the OPN-KI mice covering inflammation, oncology and fibrosis showing OPN is a potential therapeutic target over a broad range of indications. Key accomplishments- Designed, screened and generated 3 mAB candidates for pivotal mouse studies in inflammation, wound healing and inflammation that (1) Block thrombin activation and (2) Neutralize activated N-terminal OPNs- Advanced one mAB to a lead compound, humanized and assessed for expression in stable pools (ATUM) and developability assessment- Developed specific mABs for biomarker and potency assays for capture and detection of OPN species in mouse, cynomolgus and human- Generated tool recombinant proteins for screening, biomarker and potency assays
