Engineered a conditional knockdown system for a histone methyltransferase gene in Plasmodium falciparum using double cross homologous recombination.• Performed PCR, to amplify the fragments between which the gene cassette will be introduced. Western blot to asses methylation status for the extracted histones from the pelleted parasite cells.• Isolated plasmids, conducted sgRNA annealing and Gibson cloning to enable Cas9-mediated gene cassette integration• Monitored parasite developmental stages through regular blood smears to ensure experimental accuracy.
