About Stuart Ember
Protein chemist with >10 years of postgraduate (Ph.D.) experience and a track record of accomplishments in large and small molecule drug discovery and development across oncology, immunology, and infectious disease therapeutic areas. Expertise in X-ray crystallography, structure-based drug design, multi-parametric drug optimization, computational chemistry, cell biology, and antibody characterization. X-ray crystallography (crystal setup, data collection & reduction, model building, and refinement) Macromolecular modeling (PyMOL, MOE, Schrodinger, Autodock 4/Vina) Protein expression and purification (vector design, sub-cloning, AKTA FPLC affinity/IEX/SEC) Cell biology (flow cytometry, cell line generation, microscopy, reporter gene assays, proliferation assays) Antibody characterization (SE-UPLC, CE-UPLC, LC-MS/MS, AUC, Halo, MDI, HIAC, ce-SDS) Biophysical assays (nDSF, DLS, ITC, SPR, FP, FRET, HTRF, AlphaScreen) Liquid handling automation (TTP Labtech Mosquito, Hamilton, Labcyte Echo, Unchained Labs Grunt) Computational chemistry (HTS hit triage, machine learning, cheminformatics)
Experience
Structural Biology (X-ray Crystallography) - Structural & Protein Sciences
Jan 2022 — Present · Spring House, PA, US
Structural biology lead providing overall support across the small and large molecule J&J discovery portfolios. Structurally-enabled four small molecule programs across oncology, immunology, and cardiovascular therapeutic areas within the last twelve months. Determined crystal structures of Fab-antigen complexes to enable epitope mapping in support of patent differentiation efforts, as well as rational design of novel antibody-based biologics. Closely work with Chemistry and Computational Chemistry colleagues to generate analogs with improved potency and selectivity. Design protein constructs in partnership with Protein Sciences and Screening teams to enable efficient transition into the structural biology workflow.
