Studying co-translational folding pathway of a-tryptophan synthase (aTS), a TIM-barrel protein with early forming N-terminal structure- Designing primers, cloning, and expressing aTS in E.coli, followed by His-tag affinity purification and size-exclusion chromatography to isolate ribosome-nascent chain and refolded protein states- HDX-MS and single molecule optical tweezers to probe in-vitro folding on the ribosome vs protein refolding pathway.
