Current research focus on leveraging automated robotic microscopy and deep learning to model and predict cell fate under pathological stress in neurodegenerative diseases and cancer.Understanding Neurodegeneration Diseases:1. Developed nuclear biomarkers and deep learning models to predict neuron cell death2. CRISPR Screening of Parkinson\'s Disease Targets: Led a genome-wide CRISPR screen to identify survival modulators in Parkin-deficient neurons.3. Understanding Tau Pathology in Neurodegeneration: Leveraged longitudinal imaging and deep learning to identify Tau-dependent cellular dynamics; Engineered fluorescent biosensors to study Tau turnover and neurotoxicity, and enable real-time tracking of neurodegenerative pathways; Use biosensor-integrated longitudinal imaging datasets and deep learning to understand the sub-cellular changes after Tau seeding in neuron, uncovering mechanistic insights into neurodegeneration.4. Down Syndrome (DS): Characterize AD-related phenotypes using DS patient iPSC-derived neurons and organoids models; Generate stable CRISPRi iPSC lines for mechanistic validation.Technology Development- Co-developed a next-generation longitudinal imaging platform enabling light-based recording, perturbation, and isolation of individual cells, reducing inter-well variability and enabling precise optogenetic control- Integrating deep learning models to automate cell selection and analysis, creating a scalable framework for multi-modality projects- Understanding cancer drug resistance in cancer cells: Discovered biomarker for drug-resistant NSCLC, generated large dataset to implement deep learning to predict drug resistant cells prior to the drug treatment, developed methods to isolate drug-resistant cancer cells at single cell level, enabling unbiased cell selection strategies and multi-omics methods to understand the underlying mechanism of cancer cell drug resistance.
