   Antibody-based immunotherapies based on novel targets discovered from patients\' anti-tumor responses. ATRC-101: Helped develop a first-in-class antibody targeting a novel ribonucleoprotein complex and exhibiting driver antigen engagement mechanism of action. Currently in a Phase 1b clinical trial. ATRC-301: Helped generate an EphA2-targeting ADC against a novel and differentiated epitope. Achieved significant potency enhancement through lead optimization while improving developability and maintaining tumor selectivity. Currently in IND-enabling studies. Bispecifics strategy: Managing our portfolio of cell engagers and immune agonists. CD3 bispecifics: Leading ongoing >3-yr Xencor collaboration pairing XmAb with Atreca novel targets. 4-1BB bispecific platform: Widened therapeutic window by limiting 4-1BB activation to the tumor microenvironment. Demonstrated significant tumor growth regression while not damaging the liver. Target identification: Identify, validate, and weaponize novel targets spanning multiple classes (e.g, transmembrane proteins, RNA-binding proteins, oncofetal proteins, and glycans). Epitope identification: Determine novel epitopes (e.g, membrane-proximal EphA2/ATRC-301). Antibody optimization: Simultaneously improve developability, enhance potency, and maintain tumor selectivity. Employ rational design and high-throughput yeast display. Fv & Fc engineering. Responsible for: Molecular Biology, Target ID, Epitope ID, & Protein Engineering groups (>25 FTE).
