The major aims of my current research and enterprise activities are translation of the knowledge we have about Intramural Periarterial Drainage (IPAD) pathways into therapies that facilitate the elimination of Aβ and fluid for the prevention and treatment of ADAlzheimer’s DiseaseAlzheimer’s disease (AD) affects 1 million people in Britain with devastating consequences for patients and carers. Current treatments have little or no effect on the progression of the disease. One of the major causes of Alzheimer’s disease is the age-related failure of elimination of a small protein, amyloid-β (Aβ) that results in its accumulation in the brain and in the walls of arteries supplying the brain. Intramural Periarterial Drainage (IPAD) of soluble Aβ from the brainPublished work from my group has shown that soluble waste substances, including Aβ drain from the brain along thin protein layers (basement membranes) within the walls of capillaries and arteries, a process named Intramural Perisrterial Drainage (IPAD). With advancing age and in Alzheimer’s disease, elimination of Aβ fails and is deposited within basement membranes further impeding the elimination of Aβ and waste material from the brain. In addition to age, possession of the apoE4 genotype and high levels of cholesterol in the blood are major risk factors for Alzheimer’s disease. Within my group we have demonstrated that these risk factors impede elimination of Aβ and other soluble metabolites from the brain and ultimately, results in loss of homeostasis of the neuronal environment, neuronal dysfunction and cognitive decline. I am leading the design of prevention measures and therapeutic interventions that facilitate the removal of Abeta and toxic metabolites from the ageing and Alzheimer brain, preventing thus dementia.The drainage pathways from the brain extend into the nose and facilitate the clearance of proteins from the brain into the nasal mucosa.
