About Ritu Devkota

I am a Master\'s student majoring in Microbiology at Clemson University. My expected graduation date is May 2024. Currently, I am working on my Master’s thesis, where I study a specific long non-coding RNA in a human fungal pathogen, Aspergillus fumigatus. I aim to understand the role of this non-coding RNA in stress response and pathogenesis. This project has allowed me to develop expertise in gene editing techniques like CRISPR/Cas-9, which I use for creating deletion and complement mutants, and molecular cloning for generating overexpression vectors. A crucial part of my work involves extracting and purifying nucleic acids and preparing them for downstream processes like PCR, qPCR, and sequencing. Additionally, I determine the role of this non-coding RNA in pathogenesis by comparing fungal burdens in the lungs of mice infected with wild-type versus mutant strains, employing TaqMan chemistry for absolute quantification of gene targets.
