About Raashed Raziuddin
Driven by the pursuit of translating preclinical ideas into clinical candidates, Raashed is currently applying his education in biological engineering and pharmacology to develop nanoparticle formulations to deliver drugs selectively to tumor targets in the brain and elsewhere. Previously, he worked on developing anaerobic phase fermentation processes for monoclonal microbials with Evelo biosciences as immunomodulatory clinical candidates. With his degree in biological engineering at Cornell University and various lab projects in synthetic biology with Ron Weiss at MIT, Type I CRISPR with Ailong Ke, RNAi with Julius Lucks, glycoengineering with Matthew DeLisa, and flagellar microscopy with George Witman, Raashed has experience engineering biological systems at the cellular and subcellular scale for drug development.Currently, Raashed is expanding his tumor microenvironment knowledge and skillset to include the intricacies of cellular crosstalk. Between identifying and targeting protein targets specific to tumor vasculature and identifying interactions between immune players like neutrophils with endothelium and tumor cells, Raashed has expanded his knowledge in immunofluorescence, flow cytometry, and spatial transcriptomics to further develop P-selectin targeting nanoparticles.

Working in Dr. Daniel Heller’s Cancer Nanomedicine Lab at Memorial Sloan Kettering Cancer Center to study and use the induction of P-selectin after standard of care radiation to target drug delivery of chemotherapeutics for brain cancers and metastases. To this end, we are developing fucoidan-wrapped nanoparticles to encapsulate large PROTAC degraders to cross the blood brain barrier selectively into primary and metastatic brain tumors.Along with physical characterizations of nanoparticles by dynamic light scattering and atomic force microscopy, biological assays like multi-color flow, western blots, and plate-based viability and immunoassays helped uncover increased potency of nanoparticles in vitro. Throughout my PhD dissertation, I have utilized bioconjugation of PET emitters like 89Zr to fucoidan and the FDA-approved P-selectin antibody, crizanlizumab, to study biodistribution. I have developed and maintained complex in vivo models including transgenic models of sonic hedgehog driven medulloblastoma, subcutaneous tumor models of transgenic human P-selectin mice, and intracardiac injection of brain metastatic breast cancer cells. I have also refurbished and reinstated the use of a triple quadripole tandem LC-MS/MS (Shimadzu 8040) to independently quantify drug biodistribution.
