At present I am establishing a Patient Derived Tumour Fragment (PDTF) assay which involves 3D culture maintaining native tumour microenvironment, treatment with therapeutics and analysis of T cell activation/exhaustion status using flow cytometry and analysis of treatment induced secretory changes using Luminex platform.The PDTF assay is aimed at predicting patient response to therapeutics in clinic to tailor treatment as well as development of biomarkers of treatment response.Additionally, the assay is utilized for testing cancer treating compounds from a pharmaceutical collaboration.I am also working on assessing changes in TCR clones in response to Immune Checkpoint Inhibitors in non-small cell and small cell lung cancer. I am sequencing TCRs using the Oncomine TCR-beta LR assay on the Ion Torrent to examine the clonal changes. This project is a collaboration with Thermofisher Scientific.
