I am Prabhakar Pawar and I have submitted my thesis in chemical biology under the supervision of Prof. Krishna N. Ganesh at the Indian Institute of Science Education and Research (IISER) Pune, India. My doctoral thesis is focused on the Design and Synthesis of PNA and Polypeptide Mimic Nucleases. We have studied the metal complexation and nuclease activity of imidazole-modified PNA. For this, imidazole acetic acid was conjugated at various positions on the PNA. While synthesizing these PNA monomers, we came across various reactions. Some of the important reactions being hydrogenation, RaNi/H2 reduction, coupling reaction using various coupling reagents, various functional group protection and deprotection reactions, alkylation and acylation reactions. The modified PNA monomers were incorporated in control aeg PNA system at desired position by solid phase PNA synthesis protocol using Boc-chemistry and Fmoc-chemistry. The purification of PNA oligomers was carried out by reverse phase-HPLC and their integrity was confirmed by MALDI-TOF mass spectrometry. The PNA oligomers were evaluated using various biophysical techniques like temperature dependent UV-Vis absorption spectroscopy (UV-Tm), and circular dichroism (CD). The imidazole modified PNA showed interesting hydrogen bonding properties with complementary DNA and PNA.
