Built an E. coli strain engineering platform enabling rapid genome editing (~1 edit / 2 weeks) and development of multi-edit production strains (>10 genomic modifications) through pathway optimization Drove end-to-end enzyme development across 8 DBTL cycles, from library design and screening to downstream processing and lyophilization; resolved purification bottlenecks and optimized reaction conditions to achieve 90% conversion of 100 g/L substrate within 24 hours Partnered with bioprocess development teams supporting 5, 10, and 30 L bioreactors to understand scale-up constraints; redesigned screening conditions to better predict ambr250 performance, enabling reliable strain ranking and reducing scale-up risk Rescued a stalled E. coli genome-integration program by designing a two-step chromosomal integration strategy that achieved genomic expression levels comparable to plasmid-based strains while maintaining stable genome integration Led a rapid E. coli chassis-strain build under aggressive timelines, redesigning a low-efficiency genome-editing workflow and completing multiple rounds of edits within ~6 weeks to deliver strains ready for library screening Redesigned a failing Pichia expression system through end-to-end optimization of host selection, promoter engineering, media conditions, and cell lysis workflows, enabling consistent production of active enzymes Established a custom Gibson assembly protocol enabling high-efficiency assembly of ultra-large DNA constructs (50–70 kb,>10 fragments), increasing success rates from <5% to over 80% and achieving near 100% cloning efficiency in routine applications Developed a set of engineered Pseudomonas promoters spanning a wide range of expression strengths, enabling tuning of gene expression and refactoring of biosynthetic gene clusters Designed and deployed enzyme and RNA-based assays enabling high-throughput screening and cross-site tech transfer
