Analyzed proteins, oligonucleotides (ASO), and small molecules in gRED (Genentech Research and Early Development) using mass spectrometry (LC-MS,) and other analytical tools (e.g. CE-SDS, various HPLC, SEC) both in vitro and from complex biological matrices (ex vivo/in vivo). • Led molecular characterization efforts for numerous biotherapeutics (Subject Expert); discovered unpredicted amino acid modifications and physical changes (clipping/aggregation) in vitro, ex vivo, and in vivo (including many that affect biological activity) and suggested changes, leading to redesign or triage of lead molecules. • Managed collaborations, activities, and communications with outside research organizations for bioanalysis and lot testing (CROs, universities). • Independently represented department at team meetings (served as MS analytical lead), and at presentations both outside department and outside company. • Supported development of small molecule, peptide, antibody, bispecific antibody, and antibody drug conjugate (ADC) therapeutics through creation of and use of stability, functional, and binding assays; performed associated molecular characterization. • Supported development of novel therapeutics with PK/PD data generation and analysis. Successfully advanced projects from early to late stage research with aggressive timelines (e.g. transferred knowledge and assays to clinical development group) • Contributed to molecular design of drugs by determining degradation pathway and hence ways to improve stability (e.g. through substitution of unstable residues). • Developed separations methods (including automation) for bioanalysis, including ones based on LLE, SPE, immunoaffinity capture, gel purification, and HPLC/UPLC (IEX, HILIC, SEC, etc). • Performed absolute (e.g. AQUA and SISCAPA) and relative quantitative analysis (e.g. TMT); also developed top down quantitation on the proteome level (external talk at international conference).
