ABOUT PAWEL KALINSKI

My administrative efforts at Roswell focus on promoting partnerships between our clinical and laboratory faculty, biotech and pharma, in the development of new immunotherapies and other forms of cancer therapy. The research of my group aims to develop more effective cancer immunotherapies which selectively induce cancer-specific effector- and memory-type CTLs and Th1 cells, and promote their local killer functions within tumor tissues. We developed several new methods of using ex vivo-educated dendritic cells (type-1-polarized DCs, induced in the conditions which mimic viral infections) to selectively induce anti-cancer effector cells, without amplifying tumor-promoting forms of immunity in cancer patients (suppressive Tregs & MDSCs). We have also developed different types of combinatorial adjuvants to modulate DC functions in vivo and amplify the induction phase of anti-cancer immunity, and to promote selective entry of effector cells to tumor lesions. Our current projects include: • Development and clinical evaluation of different subsets of “polarized” DCs for use as cell-based therapeutic vaccines • Development and clinical evaluation of ex vivo-induced polyclonal CTLs for adoptive T cell therapies • Development and clinical evaluation of combinatorial modulators of tumor microenvironment to enhance intratumoral CTL entry and modulate intratumoral expression of PDL1, PDL2 and other checkpoint ligands • Development of combinatorial adjuvants to induce DC polarization in vivo • Interplay between immune system, chemo- and radiotherapy, and psychologic stress • Counteracting cancer-related immunosuppressive mechanisms and their amploification in transplantation and autoimmunity The resulting paradigms are being currently tested in phase I/II and phase II clinical trials in patients with different forms of cancer cancer. We are also developing similar treatments for patients with premalignant lesions and chronic infections which are resistant to standard forms of treatment.
