I am currently working as an Undergraduate Research Assistant in the Kielkopf Lab at the URMC. I am starting a project where I will investigate the role of reactive oxygen species (ROS) in regulating PPM1D phosphatase. PPM1D mutations or overexpression suppress DNA damage responses and p53-mediated apoptosis, promoting chemoresistance. It is hypothesized that ROS-sensing cysteine residues uniquely positioned in PPM1D modulate its activity through redox-dependent structural changes. My experiment will first characterize how oxidation alters PPM1D’s enzymatic function and cellular stability using biochemical assays, cysteine-mimetic mutants, and redox proteomics. After this, I will then screen covalent small-molecule libraries targeting these cysteines, which could be used to develop selective PPM1D inhibitors. These hits will also be validated in orthogonal cell-based models measuring p53 activation, substrate dephosphorylation, and chemosensitization.

ABOUT NATASHA SIECZKIEWICZ

My name is Natasha Sieczkiewicz, and I am student at the University of Rochester currently pursuing a BS in Molecular Genetics and a minor in Anthropology. I am passionate about understanding the complexities behind molecular mechanisms and how this understanding can be applied to studies regarding genetic diseases and conditions. After graduating from the University of Rochester, I aim to work in a research laboratory and gain experience before ultimately pursing a PhD in genetics, and eventually work for a company where my research could be used to enhance the lives of others.I currently work as an undergraduate researcher in the Kielkopf lab at the URMC. Beyond academics, I am drawn to other learning experiences that strengthen my academic goals. I have worked as an undergraduate TA for a genetics course and another introductory biology course focusing on ecology and evolution. Additionally, my past employment experiences have equipped me with the skills I need to succeed in new positions and the mindset to strive to understand the world around me.
