Department of Basic Science, Fred Hutch Cancer CenterAdvisor: Dr. Sue BigginsTopic: “Regulation of kinetochore-microtubule attachments”• Generated mutations in cells utilizing CRISPR and homologous recombination on a critical protein required in eukaryotic cell division. Optimized the protocol for purification of the protein in the lab which is now followed by the lab members. The mutation turned out to be essential for the cell division as it perturbed the localization of major proteins at the kinetochore.• Optimized fluorescence microscopy to execute live cell imaging in Delta vision microscope to observe the localization of different proteins under the mutant condition.• Experienced in in vitro protein purification of His tagged protein by IMAC, bead-based immunoprecipitation of 3FLAG or 3V5 or 13Myc tagged proteins and subjected to bind different proteins of the kinetochore. The binding assay resulted in previously unknown binding phenomenon of the two cell division proteins, as observed from silver staining, SDS PAGE and western blot. Utilized Alphafold and Pymol to determine the binding beforehand.• Experienced in optical trapping assay using functionalized polystyrene beads conjugated to proteins and reconstituting in vitro single molecule protein-protein interaction by laser trap. • Trained and mentored undergraduates and junior graduate students to successfully execute and document experiments, helping them to become successful scientists in future. Prepared lab reports, and SOPs and updating technical protocols of the lab.
