Research Projects:1. Defining the changes in molecular profile of the aging intervertebral disc.• Used bulk tissue and single-cell RNA sequencing to understand the changes in molecular profiles and identify key immune cell populations that contribute to tissue degeneration with aging.• Successfully optimized a method to isolate high quality RNA from rat and mouse intervertebral disc for RNA sequencing.• Proficiency in using R packages to analyze bulk and single cell RNA sequencing datasets.2. Understanding the role of systemic and local tissue inflammation during intervertebral disc degeneration.• Experience in working with inducible chronic inflammation mouse model, immune receptor knock-in/knock-out model, and conditional KO model of danger associated molecular patterns.• Lead in testing various biologics (small molecule inhibitors, neutralizing antibodies) for immune cell recruitment and activation.• Experience in isolating PBMCs and characterizing macrophage polarization state using flow cytometry.• Experience in quantifying blood serum cytokine levels in sepsis mouse model.• Experience in ex vivo, in vitro co-culture system to study macrophage polarization and migration.3. Delineating the crosstalk between intervertebral disc degeneration and peripheral nervous system.• Experience in dorsal root ganglion (DRG) isolation and retrograde neuronal tracing in both mouse and rat models.Mentorship and Training:• Mentored 10 trainees and staffs at various levels thus far (undergraduate, graduate, laboratory technician).

At Columbia University, my research focuses on defining the molecular profile of aging intervertebral discs. Utilizing RNA sequencing I am interested in unravelling immune cell contributions to tissue degeneration. With proficiency in R for data analysis, I have optimized methods for high-quality RNA isolation from rodent models, supporting our insights into systemic and local tissue inflammation\'s role in disc degeneration.As an educator, my role entailed directing a Dental Pharmacology course, where we implemented creative communication and active learning strategies. This experience, coupled with our current research, reinforces my commitment to advancing musculoskeletal health through cutting-edge scientific inquiry and transformative education.
