Experimental PhD skills-MICROBIOLOGY: engineering of bacteria, OMVs isolation and purifications, chemically competent cells preparation, bacterial cultutres, Plasmids construction using the Polymerase Incomplete Primer Extension (PIPE)-IMMUNOLOGY: analysis and optimization of adaptive immune responses, ELISA, ELISpot, Western blotting. Splenocytes, lymph node cells and PBMC isolations from mice and coulture of these cells-IN VIVO EXPERIMENTS: intramuscular, subcutaneous and intraperitoneal immunizations. Mouse surgery. Experimental preDOC skills- TECHNICAL SKILLS IN CELL BIOLOGY, Primary cell culture (PBMC and MSC derived from human bone marrow, and MSC from pig’s bone marrow). Cell lines. MSC, PBMC and CD34+ cells isolation by peripheral/marrow blood with ficoll. Co-culture. Differentiation of MSC in osteoblasts and adipocytes to determine “stem potential”. Freeze cells. MSC seeding in oesophageal scaffold-TECHNICAL SKILLS IN MOLECULAR BIOLOGY: RNA isolation, PCR, cDNA synthesis, quantification and analysis of the RNA integrity, real time RT-PCR- TECHNICAL SKILLS IN HISTOLOGY AND MICROSCOPY: light microscopy, image analysis.
