About Kyle Chamberlain
I am a Senior Scientist and virologist with over 15 years of experience advancing AAV gene therapy, viral vector engineering, and translational research across academia and biotech. My work spans early discovery through IND-enabling biodistribution, analytical development, and preclinical translation, with programs supporting both cardiovascular and CNS indications. My technical foundation includes ddPCR/RT-ddPCR assay development, viral genome and expression quantification, NHP/mouse tissue workflows, and analytical method qualification for first-in-human programs. I’ve designed and evaluated vectors carrying traditional gene therapy payloads, as well as regulatory oligonucleotide payloads including miRNA, siRNA, and other small RNA-based modulators, supporting studies focused on target suppression, transgene regulation, and tissue-specific expression. At Affinia, I lead analytical workflows for biodistribution and expression studies, manage CRO partnerships, troubleshoot complex assays, and generate IND-supportive data packages and regulatory documentation. My work directly contributes to dose selection, vector optimization, and the translational strategy for AAV therapeutics.I’m driven by bringing rigorous molecular virology, assay innovation, and cross-functional collaboration together to accelerate the next generation of AAV therapies for CNS, cardiovascular, and other high-impact disease areas.Specialties:AAV gene therapy • Viral vector engineering • CNS & cardiovascular programs • ddPCR/RT-ddPCR • Biodistribution • Oligo payloads (miRNA, siRNA) • Translational science • IND-enabling studies • CRO management • Molecular cloning • Oncolytic virology • High-containment virology
Experience
Senior Scientist
Jul 2024 — Present · Waltham, MA, US
Lead scientist for ddPCR/RT-ddPCR assay development, biodistribution studies, and vector expression quantification supporting AAV programs for CNS and cardiovascular indications.• Design, optimize, and execute analytical workflows for mouse, NHP, and human tissues; responsible for end-to-end sample processing, nucleic acid QC, and data integrity.• Support programs using traditional gene therapy payloads as well as modulatory oligo payloads (miRNA, siRNA) enabling targeted regulation and suppression strategies.• Manage external CRO partnerships for preclinical NHP studies, analytical method transfer, and development of clinical-grade ddPCR/RT-ddPCR assays.• Generate and review IND-supportive data packages, contributing to dose projections, biodistribution assessments, and regulatory documentation for first-in-human programs.• Implement internal QA processes enabling selected in-house studies to meet regulatory expectations.• Regularly present translational findings to leadership, informing vector engineering, dose selection, and program strategy.
