ABOUT JOSEPH CANDIELLO

in Bioengineering with a concentration of biophysics and cell biology. The focus of this work was using a combination of atomic force microscopy and biological assays to investigate the important biomechanical properties of extra-cellular basement membranes with a focus on aging and disease pathogenesis. Undergraduate work was in Engineering Science and Mechanics at the Pennsylvania State University.Post-Doctoral WorkStem cells receive both chemical and mechanical cues from their environment which helps drive differentiation. Current work focuses on engineering and applying novel platforms to take advantage of immediate microenvironment cues to tailor systems to maximize the response of embryonic stem cells (ESCs) towards a desired end cell phenotype. This includes whether specific organic or inorganic materials and nano-materials have a biophysical effect on downstream ESC lineage fate, which may be used as part of a larger multi-faceted differentiation approach.Beyond just passive environmental stimulation, focus is on investigating whether multicellular 3D cell culture systems, called organoids, are a more suitable platform to reach our objectives. Organoids require the development and combination of (1) multiple organoid specific cellular components including functional paranchyme,(2) a platform for generating and organizing 3D tissue structures, and (3) development of supporting organ specific vasculature and supporting mesenchyme. Therefore, novel strategies to engineer organoids and their specific vascular systems within 3D tissue constructs to support downstream therapeutic applications as well as organ-on-a chip applications are being pursued.Currently Product Manager at RoosterBio, Inc.
