About John W Rumsey

I lead GMP Business Operations at the Duke Human Vaccine Institute (DHVI), directing a multidisciplinary team responsible for environmental monitoring, equipment lifecycle management, facilities operations, supply chain, procurement, and GMP warehousing within a clinical-stage vaccine manufacturing environment. I am accountable for operational readiness, regulatory compliance, financial stewardship, and performance optimization across DHVI’s cGMP facilities. DHVI develops and manufactures protein nanoparticle and mRNA-based vaccines targeting HIV, influenza, SARS-CoV-2, and emerging infectious diseases, translating academic innovation into compliant manufacturing processes that support Phase I clinical trials and early-stage immunotherapy programs. My leadership integrates financial strategy with data-driven operational execution in highly regulated environments. I oversee budgeting, forecasting, and business modeling to enable sustainable growth and disciplined cost management. Through predictive modeling, structured analytics, and scenario planning, I drive optimization of preventive maintenance and environmental monitoring programs—improving efficiency while safeguarding compliance and risk integrity. A central focus of my work is translating complex operational data into risk-informed decisions that align cost, performance, and regulatory expectations. I apply structured, risk-based frameworks to strengthen asset reliability, optimize monitoring strategy, and ensure that operational effort is proportionate to system criticality and organizational priorities. This approach advances both near-term execution and long-term strategic resilience in GMP manufacturing systems. I hold a Ph.D. in biomedical sciences with a research background in tissue engineering and neuromuscular biology. My academic work centered on advanced microphysiological systems and defined in vitro platforms designed to model complex biological interactions with translational relevance. Across my career, I have partnered with academic, pharmaceutical, and government stakeholders to convert scientific and regulatory complexity into scalable, performance-driven operational systems. Core strengths include GMP operations leadership, financial and operational analytics, risk-based maintenance strategy, environmental monitoring optimization, supply chain oversight, and translational program execution.
