Utilized 10x single-cell genomics to characterize RA antigen-T cells. Investigated protein translation in T effector cells with regard to Treg suppression. Identified Tenascin-C as a potential cause for rheumatoid arthritis (RA), providing insights for RA development and treatment. Identified disease-specific T cell receptors (TCR) through single-cell RNA sequencing of PBMCs from RA patients, followed by re-expression and transduction of these TCRs into human CD4 T cells for further functional assays. Employed CRISPR-CAS9 and AAVs to knock out or knock-in genes in human CD4 T cells, enabling the study of autoimmune diseases and conducting immuno-function assays. Discovered a drug candidate, AD2900, and investigated its impact on lymphocyte trafficking in human PBMCs and a mice EAE model (multiple sclerosis). Characterized the functional switch of LysRS, an enzyme critical in tRNA structure change. Uncovered enhanced effectiveness of commercial anti-cancer drugs through co-administration with sphingolipid analogs, regulating ceramide metabolism. These experiences showcase a diverse skill set and research expertise in immunology, with a focus on autoimmune diseases, immuno-oncology, and biomarker development.
