Conducted data visualization and interpretation for integration site analysis (ISA) results of anti-CD33 CAR T drug products, drafted and finalized ISA report for the IND Application, and addressed FDA inquiries regarding the report.• Analyzed multimodal single-cell sequencing data of CD33 CAR T drug products and matched starting T cell materials from the same donors, with/without dual mobilization, to assess mobilization impact on T cell populations and evaluate the starting material options for treatment system of clinical trial.• Conducted multimodal single-cell analysis of AML PDX tumors to characterize tumor heterogeneity and interpret responses to CD33 CAR T treatment in PDX-engrafted mice.• Performed analysis on NGS (PacBio and Illumina) data from screening of pooled CAR libraries to enable high-throughput evaluation of approximately 1,600 CAR design variants and identify novel CAR constructs with enhanced persistence.• Visualized and interpreted ISA and bulk TCR-Seq data from cytokine-independent growth samples to assess clonal expansion of CD33 CAR T drug product.• Led translational research on Trem-cel, an allogeneic CRISPR/Cas9-edited HSPCs therapy product with CD33 knockout, by analyzing gene editing and bulk TCR/BCR sequencing data to evaluate lineage-specific CD33 knockout and characterize T/B cell immune reconstitution and clonal diversity.• Analyzed Mission Bio single-cell DNA and protein sequencing data from edited HSPC-engrafted mouse bone marrow to determine genotypes across different cell populations.• Performed in-depth analysis on amplicon sequencing data from genome editing technologies, including CRISPR/Cas9-mediated gene knockout, ssODN-based HDR editing, ABE and CBE, to support development and optimization of various preclinical assays.• Applied rhAmp-Seq and custom bioinformatics pipelines to evaluate translocation potential in multiplex-edited HSPCs when targeting the CD33 and CLL1 genes for knockout.
