  1. Principal Scientist, Analytical Development and Characterization

    Alnylam Pharmaceuticals

    Jan 2021 — Present

Lead the technical deliverables and activities for achieving project milestones in matrix teams comprising of AD, QC, QA, regulatory CMC, and program teams. • Developed, qualified, transferred, and validated multiple LC-MS and GC-MS based CMC assays (impurity characterization, sequence confirmation, structure confirmation, genotoxic impurities) for novel siRNA therapeutics and their starting materials from early stage to post-commercial stages. • Lead matrix teams which performed method readiness assessment and transfer to multiple testing sites (internal QC and CMO/CTL) in parallel to support in process control, release testing, and purity/impurity methods for raw materials, intermediates, and excipients. • Conducted stability forced degradation characterization and genotoxic impurity assays supporting process development and QC release testing of cGMP manufacture from pre-clinical to commercial stages. • Advanced the state of the art in oligonucleotide impurity characterization (GC, LC, SFC) by directing capital expenditures and developing novel strategic separation technologies resulting in industry leading technical capabilities including: Multiple Custom LC-MS purification platforms at multiple scales to automate various impurity characterization workflows for oligonucleotides and their formulations (including conjugates and lipid nanoparticles) in support of non-GMP and GMP activities. Conceptualized, refined, and implemented a multi-dimensional impurity characterization platform that significantly increased the efficiency (>3x faster) and sensitivity (>3 logs) of the impurity characterization workflow. This strategic capability results in richer data sets, expanded impurity characterization capacity, and achieves this without increasing headcount.
