multimmune GmbH is a private, clinical-stage biopharmaceutical company which has developed a unique and proprietary technology platform on the basis of the founding scientists\'(Prof. Em. Gabriele Multhoff, Dr Claus Botzler) discovery that cell surface-bound Hsp70 is uniquely expressed on a large proportion of cancers (~50-75%), but not on healthy cells. This makes this form of Hsp70 an excellent and broadly-applicable molecule on which to base the development of innovative therapeutics and diagnostics (\'theranostics\').A Phase I clinical trial of our first generation, non-expanded TKD/IL-2 activated autologous NK cell therapy (ENKASTIM™) involving patients with metastatic colorectal cancer and non-small lung cancer (NSCLC) after standard of care (SOC) in Germany demonstrated safety and signs of efficacy. A Phase IIa (randomized, controlled) clinical trial of ENKASTIM™ in patients with advanced (Stage inoperable NSCLC conducted in Germany demonstrated safety, promising clinical benefit and survival.Tumour cells expressing membrane Hsp70 actively release exosome-like lipid vesicles expressing Hsp70 on the surface of the vesicles into the blood. We have developed a novel sandwich ELISA for quantifying free and exosomal Hsp70 in the serum of patients with different tumor types. The Hsp70-exo ELISA is the only non-invasive ‘liquid biopsy’ blood test which can be used to detect the presence of a wide range of cancers, even in their early stages, and to prognosticate and monitor therapeutic response and/or disease recurrence in multiple cancer settings. The Hsp70-exo ELISA is a patented (US granted, EPO pending) CE Marked test. Alphageneron Pharmaceuticals Inc. USA and multimmune GmbH, Germany signed an Exclusive Worldwide License, and an Option to Purchase Agreement for its membrane Hsp70-based immunotherapy and diagnostics platform technology, in 2020.

ABOUT A. GRAHAM POCKLEY

I am the CEO of multimmune GmbH, Munich, Professor Emeritus of Immunobiology (Nottingham Trent University) and Chief Scientific Officer of Alphageneron Pharmaceuticals Inc. USA, Radiance Biopharma Inc. USA and ExoCellular Diagnostics Inc. USA. I am also the former Director of the John van Geest Cancer Research Centre at Nottingham Trent University, UK (20••••20), and the Founder and CEO of the flow cytometry resource Chromocyte, the assets of which were sold to Biocompare. multimmune GmbH is a private, clinical-stage biopharmaceutical company which has developed a unique and proprietary technology platform on the basis of the founding scientists\' (Prof. Em. Gabriele Multhoff, Dr Claus Botzler) discovery that cell surface-bound Hsp70 is uniquely expressed on a large proportion of cancers (~50-75%), but not on healthy cells. This form of Hsp70 is an excellent and broadly-applicable molecule on which to base the development of innovative therapeutics and diagnostics (\'theranostics\'). Based on its discovery of membrane Hsp70, multimmune has succeeded in developing a powerful pipeline of cancer theranostics. ENKASTIM allows, for the first time, a unique and specific activation of human natural killer (NK) cells which are programmed to recognise and kill the large proportion of cancers expressing membrane Hsp70. These cells destroy tumors and metastases that are invisible to cytolytic T cell immunotherapies. This proprietary autologous natural killer (NK) cell therapy targeting membrane Hsp70 has successfully demonstrated safety and delivered signs of efficacy in completed Phase I and Phase II clinical trials in Germany. Membrane Hsp70-expressing tumors can also be imaged and targeted using theranostics based on a unique monoclonal antibody which can detect the membrane form of Hsp70 and a patented peptide which binds to membrane Hsp70. multimmune\'s lead diagnostic candidate is the patented and CE-marked Hsp70-exo enzyme immunoassay (ELISA) which detects lipid-associated Hsp70 in the peripheral circulation, levels of which can be used to detect the presence of tumors expressing membrane Hsp70 and prognosticate disease, as well as monitor therapeutic response and disease recurrence. My career has focused on the immunobiology of disease, the development of new immunotherapeutic strategies for treating aggressive and \'orphan\' cancers, new approaches for analysing and interpreting complex datasets and defining new biomarkers of disease presence, status and response to therapy in patients with cancer.
