Hit Discovery and Optimization PFIZER (2018-present) • Lead Customized Solubility/Stability Assessment team in support of early formulation selection -Molecular Properties Group (20••••18) • Established EPSA as industry leading SFC based IMHB and peptide permeation prediction tool. • Enabled LC-Biofractionation. Identified nanomolar contaminant responsible for activity-Autopurification - technology evaluation (20••••11) • Identified cholesterol as natural agonist of orphan receptor ROR gamma. Enabled first crystal structure of protein, providing tool for assay development. • Devised and implemented on-column solvent exchange for HPLC fractions. Reduced drying times from 18 to 2-4 hours, enabling same day sample availability for priority samples with higher quality dry down. • Drove purification cycle times from 5 days, down to 2 for over 90% of the samples submitted annually. Earned high level of Individual Performance Award recognition. • Established mentoring relationship with junior colleague, leading project that identified improved conditions for DMSO evaporation, eliminating residual solvents for >90% of samples-Analytical chemistry ID-purity-activity (20••••05) • Identified activity of lead c-MET hit to be due to minor contaminant, not target molecule. This compound lead to optimized derivative which entered FIH and FIP studies in 2008. • Provided LC-MS-Bioassay follow up and showed lead pyrazolone hit to convert in solution into a bis-pyrazolone responsible for the activity, identifying new scaffold for the kinase design team -Natural product chemistry - 2001) • Isolated novel natural product inhibitor of TF/VIIa and determined absolute structure, leading to design enabling bound protein crystal structure. • Established GC-MS methodology for metabolomics, utilizing SPME technology in effective prediction of early toxicity.
