I am developing quantitative cell-based imaging assays to directly visualize, track the uptake and intracellular fate of endotoxins such as lipopolysaccharide and lipoteichoic acid in hepatocytes in response to RNA lipid nanoparticle therapies for severe bacterial infections and sepsis. This includes assay development, experimental design, imaging, and data analysis to resolve uptake dynamics at the single-cell level in collaboration with Russel, Cullis, and Leslie Labs.A key outcome of the project is to establish an imaging-based screening platform that can be used to evaluate and optimize next-generation RNA lipid nanoparticle formulations for clinical translation in the treatment of sepsis and acute bacterial infections.
