Presently I am working at Tata Medical center, Kolkata as Scientist C under System Medicine Cluster in Cancer project. In this project I develop around 90 primary culture from tissue and body fluid of ovarian and cervical cancer patients. This primary culture further utilized for drug screening and proteomic/genomic analysis. I also develop Tissue microarray to evaluate the special distribution of immune cells and their interaction with stromal cell by utilizing Multiplexing method. I also made 4 primary cancer cell lines by immobilization method in ovarian cancer. As PARP inhibitor (Olaparib) have been approved as drug for ovarian cancer patient with homologous recombination deficient patients, so all primary cultures from ovarian cancer patients have being screened for Olaparib sensitivity and also for other drug. Cancer tissue in form of (FFPE, Snapfreez and RNALater) and primary culture are store in Bio-bank, which is leading by me. My lead project to identify the cancer associated subtypes and their regulatory activity in immune infiltration in homologous recombination stratified ovarian cancer. Here I am performing label free proteomics from FFPE and primary cells. All tumor associated primary cells (epithelial cell, fiboblastic cell, endothelial cells and immune cells) are being sorted by MACS magnetic column and by FACS for further downstream functional assay like trans well migration assay, adhesion assay etc. I also develop omentum and ovary organoid tumor model which is being utilized for drug screening and immunological functional assay. Extracellular matrix is one of the major component of tumor microenvironment and it’s composition is the major landmark for tumor fibrosis or surgical resectibility for ovarian cancer patients. This composition of the Extracellular matrix is still a major area of investigation. I also devoted myself to dissect the composition by Extracellular matrix proteomics and glycomics.
