Project aim is to assemble histone octamers on the DNA to form a nucleosome complex; mono-nucleosomes and the array as a sample for nanopore experiments. Research involves exploring, at a single-molecule level, the forces and dynamics of chromatin compaction and its modulation by architectural proteins. Performing Nucleosome-RCC1 assays to better understand the higher order structure of chromatin.CRISPR-Cas system :) CRISPR-cas9 is well studied bacterial adaptive system. Using cas9, the RNA-guided DNA endonuclease from a type II CRISPR system, we aim to design a construct mimicking the nucleosomal ‘beads on string’ structure. Using biochemical methods and the steps to purify the wildtype and the mutant(dCas9) form of cas9 in the lab, in future dCas9 will be used to design the construct of interest serving for the calibration of in-house nanopore setup to study the nucleosomal distribution profile of cells.
