About David Posson
Current work: Protein sciences in a pharmaceutical setting. Target protein expression/purification for use in binding assays, target-based screening approaches, and structure determination by cryo-electron microscopy.My career has focused on the mechanisms underlying ion channel function in biology using biophysical techniques and a structure-function approach. By understanding how these crucial proteins work, many diseases will be better managed in the future by drugs that modulate the specific activity of ion channels. Past work includes the mechanism of voltage-dependent gating in the potassium (KV) channel family and structure-function studies of gating and permeation using model prokaryotic potassium channels, including a prokaryotic calcium-activated potassium channel, MthK. Currently interested in studies of disease-relevant human ion channels such as CFTR.General techniques employed include: Channel expression and recording in Xenopus oocytes. Fluorescent labeling and Fluorescence Resonance Energy Transfer (FRET) assays of channel conformation. Channel cloning and molecular biology. Channel expression and purification from E.coli for structural and biochemical studies. Channel crystallization and structure determination using X-ray crystallography. Channel reconstitution and recording in artificial planar lipid bilayers. Channel reconstitution into apolipoprotein bound lipid discs. Channel ensemble activity assay using a stopped-flow liposomal thallium flux assay. Isothermal Titration Calorimetry (ITC) and molecular binding assays.
