Operating within the Vector Sciences team as part of Technical Operations/CMC. Upstream (USP) and downstream (DSP) processing of AAVs, as well as analysis of samples. Working on optimising the vector system such as capsid modifications, promoter screening and changes to genetic payload cassette. Techniques include- Molecular biology- Adherent and suspension (shake/bioreactor) vector production- Iodixanol ultracentrifugation, commercial methodology and AKTA concentration of vector- Depth filtration- AKTA capture (affinity) and polish (ion exchange) of vector preps- Analysis of vector prep intermediates and products: SEC-MALS, dPCR (manual and automatic), SDS, CE-GI, capsid lateral flow, endotoxin tests- Cell culture- Promoter optimisation of the transgene plasmid
