Led a collaborative project with Dr. Steven Foung\'s lab at Stanford University focused on profiling immune cell responses to experimental HCV vaccines like rAAV and mRNA studies. Designed and optimized a 32-color flow panel on the Cytek Aurora to profile B and T cells in macaque PBMCs immunized with various HCV vaccine candidates. Selected and validated monkey-reactive antibodies, including cross-reactive human clones, ensuring species compatibility through literature and vendor resources.3. Optimized antibody titrations to maximize positive/negative peak separation and improve resolution of dim markers in rare or transitional immune subsets. Cultured and maintained human embryonic kidney HEK293 and heptocellular carcinoma HUH7 cell lines for virological assays.5. Engineered HCV pseudo particles HCVpp by transfecting 293T cells using bacterial plasmid vectors that encodes E1E2 viral glycoproteins and a luciferase reporter gene.6. Treated HCVpp with postpartum patient serum to evaluate natural neutralization by broadly neutralizing antibodies (bNAbs). Analyzed bioluminescent signals to assess serum-mediated inhibition of HCV infection. Maintained sterile technique and compliance with biosafety protocols.
