Provide cell-based screening and mechanistic assay support for multiple projects in the Johnson & Johnson portfolio. Design and conduct experiments to determine mechanisms of toxicity and/or develop screening assays to de-risk unexpected findings involving cells from a variety of target organs.• Established Luminex Cytokine Assay to assess inflammatory cytokines collected from 3D stratified skin cultures and assessed cell viability using MTT and CTG assays in 2D and 3D skin models• Investigated and assisted in developing an in-vitro screening assay to measure Hand-Foot Syndrome (HFS) in stratified 3D and 2D skin cultures via MTT viability assay, Transepithelial Electrical Resistance, ROS assay, and Luminex Cytokine Assay to look at the potential roles of inflammatory cytokines with regards to HFS• Assessed cytotoxicity, inflammatory profiles, and lysosome biogenesis of lower and upper airways of the lungs when exposed to hypertension reducing compounds in stratified 3D and 2D cultures via MTT assay, Luminex Cytokine Assay, immunofluorescence on the Operetta, and WES Capillary Electrophoresis • Assisted the development of an in-vitro assay differentiating human PBMCs to mature osteoclasts to test effects of agonist compounds on the differentiation and inhibition of PBMCs into osteoclasts and subsequent measuring of mature osteoclast activity through TRAP assessment, mRNA quantification via qPCR, morphological assessment via immunofluorescence on the Operetta and Incucyte, and bone resorption activity• Oversaw and mentored intern’s work stimulating THP-1 cells with PMA to assess cytokine profiles of stimulated cells over time as well as teach basic laboratory techniques, generating and presenting data to the toxicology team, and data analysis using Belysa software, GraphPad Prism, and Microsoft Excel
