About Chad Toledo

During my graduate and post-doctoral studies, I studied Glioblastoma Multiforme (GBM), the most common and aggressive form of brain cancer. To identify new therapeutic targets for GBM, I employed functional genetic technologies, shRNA and CRISPR-Cas9, in patient-derived GBM stem-like cells (GSCs) and also primary non-transformed human neural stem cells (NSCs), non-neoplastic tissue of origin controls, to identify genes differentially require for GBM patient isolates, but which have non-essential or redundant functions in neural stem cells.The ubiquitin proteasome system is a dynamic and complex regulated network that controls the fate of cellular proteins. At Nurix, I utilized my expertise to perform target discovery and validation studies in oncology and immuno-oncology. In addition, I assessed small molecules and lead compounds that enhance or inhibit the activity of the targeted ligase.Summary of Qualifications• Ph.D. in Molecular and Cellular Biology.• Over 13 years of hands-on experience in cellular and molecular biology and 2+ years in biotechnology.• Extensive knowledge in: cancer biology, especially glioblastoma multiforme (GBM), stem cell biology, ubiquitin proteasome system and ligase biology, immune-oncology, kinetochore biology, tumor microenvironment, cell cycle, mitosis, si/shRNA, and CRISPR-Cas9.• Extremely skilled in: mammalian cell culture (including patient-derived GBM isolates, primary neural stem cells, suspension cells, among others), assay development, Meso Scale Discovery assays, viral transductions, high-throughput shRNA and CRISPR-Cas9 screening for target discovery, next generation sequencing, RNA-Seq, cell line generation, genomic engineering, viability and phenotypic assays, multi-flow cytometry, FACS, ELISAs, immunohistochemistry, and time-lapse microscopy.• Performed multiple genome-wide and focused viability screens in preclinical in vitro and in vivo GBM models for target discovery.
