In this process we cloned two genes from Candida albicans (RNT-1p and CDR-1p) in binary vector pCAMBIA- 2300. Further, it has been transferred into Nicotiana benthamiana and Nicotiana tabacum through Agrobacterium mediated transformation. Transgenic plants were confirmed by PCR as well as southern blotting. Confirmed transgenic overexpressing RNT-1p was raised till T2 generation and these transgenics were infected with Potato virus X (PVX) and Cocumber green mottle mosaic virus (CGMMV). Symptoms of infected plants were noted till 30 days post inoculation and level of infection was detected using northern blotting and ELISA. We found that symptoms were weak compared to non transgenic plants. It has been proved in-vitro in our lab that RNT-1p is capable of degrading double strand RNA molecule. Whereas fungus infection to transgenic overexpressing CDR-1p with Rhizaconia solani and Fusarium oxysporum does not show much effect with respect to wild type (N. benthamiana) infected with these fungus. In association with this, I have performed bioinformatic analyses to find simple sequence repeats (SSR) in plant-infecting viruses such as geminiviruses, potyviruses, tobamoviruses, carlaviruses and potexviruses. During this analysis, I characterize the location of these SSR within the gene of these genomes and correlate with its function.
