Advisor: Prof. Philip S LowHere, I am working on a project by designing and synthesizing new liver-specific asialoglycoprotein receptor-targeting GalNAc-drug conjugates for treating non-alcoholic steatohepatitis (NASH) and other liver diseases, and to support the patent application. 1) Development and synthesis of a liver-specific targeting ligand conjugates with a flexible and novel oligo-heterocyclic (oligo-piperidine-structural motifs) rigid linkers focusing on NASH and other liver disease treatments, gaining new expertise in solid-phase peptide synthesis.2) With the developed linker, I conjugated it with GalNAc and an NIR dye to perform the biodistribution study to assess the liver uptake in vivo in mice.3) I also synthesized the GalNAc-oligo piperidine-4-carboxylic acid rigid linker-T3 (THR β-agonist) conjugate. This was a lengthy synthesis, taking approximately 15 steps and modifying a few steps to improve the yield and purification technique compared to the previously developed method. 4) I am involved in synthesizing other piperidine rigid linkers by structural modifications with the aim of increasing the efficacy of the drug conjugate.5) I completed Purdue University\'s practical training on \"Animal Handling\" and received an animal use qualification number (Q-number) issued by the Purdue Animal Care and Use Committee (PACUC).6) I gained hands-on experience in utilizing reverse-phase HPLC and UPLC separation techniques to obtain compounds (Molecular weight > with high purity.
