The team at Radiant Biotherapeutics is developing a novel modular multi-valent and multi-specific biologic platform called the MultabodyTm (MULTi-specific, multi-Affinity antiBODY) which offers several advantages over traditional antibody therapeutics and other multi-valent platforms. The Multabody platform offers the unique opportunity to introduce up to 3 different antigen specificities while retaining avidity. This allows targeting of multiple epitopes of a single molecule or engagement of multiple targets to increase specificity, augment cell-cell interactions or cluster target molecules. The Multabody also incorporates IgG Fc fragments to achieve IgG-like PK and can be tuned to alter binding affinity to Fc gamma receptors to modulate effector function. Moreover, the Multabody platform has been designed to include several key attributes from a developability perspective:(1) the ferritin scaffold and antibody framework are human sequences minimizing the immunogenicity risk,(2) MBs self-assemble into highly stable particles with robust thermal stability characteristics, and (3) well established antibody developability processes can be utilized to identify Multabodies with favorable drug-like properties.
