Developed a high-throughput magnetic bead-based purification method for monoclonal antibodies (mAbs) and bispecifics by integrating manual protocols with Hamilton liquid handler automation. Performed antibody characterization using capillary electrophoresis: CE-SDS: mAb purity and heterogeneity CE-cIEF: Charge variant profiling CE-glycan: Glycan modification analysis Established HPLC-based methods for: Protein A (ProA): mAb concentration measurement CEX: Charge variant analysis AEX: AAV empty/full capsid characterization SEC: Purity assessment for mAbs and AAVs Built an in-house workflow for N-glycan release and HILIC UPLC profiling. Investigated acidic species in mAbs via LC-MS-based peptide mapping: Ruled out glycosylation heterogeneity Identified alternative PTMs including glycation and oxidation Developed an in-house method using CpB treatment to efficiently remove C-terminal lysine residues.
