Focused on modeling the pathophysiology of Amyotrophic Lateral Sclerosis (ALS) through the development of a cortico-spinal tract-on-a-chip system and investigating RNA dysregulation associated with neurodegeneration. • Proficient in culturing human induced pluripotent stem cells (hiPSCs) and directing their differentiation into cortical and spinal motor neurons. • Skilled in assessing neuronal maturation, activity, and synaptic connectivity using multi-electrode array (MEA) platforms. • Conducts immunofluorescence staining (IF) to validate lineage-specific differentiation and phenotype fidelity. • Introduces antisense oligonucleotides (ASOs) to induce TDP-43 knockdown and studies downstream effects on cryptic exon inclusion, RNA processing, and gene expression. • Performs RNA extraction, cDNA synthesis, quantitative PCR, and protein analysis to evaluate molecular changes associated with ALS pathology. • Skilled in interpreting large datasets using GraphPad Prism and neural metrics software for functional and molecular correlation.This work advances in vitro disease modeling of ALS and supports the development of RNA-targeted therapeutics for neurodegenerative and neuromuscular disorders.
