Actuate is a clinical stage oncology-focused drug development company that is developing elraglusib (9-ING-41) for the treatment of advanced cancer. Elraglusib has been evaluated in a large, multi-part phase 1/2 trial in over 300 adult patients and has demonstrated early signals of anticancer activity in several histologies including pancreatic, lung, cervical, endometrial, head and neck, colorectal cancer and melanoma. A randomized phase II study in patients with metastatic pancreatic cancer evaluating the combination of elraglusib plus gemcitabine and nab-paclitaxel is currently enrolling patients in the US and Europe and additional phase II trials are being planned. Elraglusib is a novel, small molecule inhibitor of GSK-3 that has demonstrated multimodal activity that includes attenuation of gene expression of antiapoptotic proteins, inhibition of damaged DNA response, and modulation of immune function including regulation of PD-1, LAG3 and TIGIT pathways and expression of TGFbeta and pro-inflammatory cytokines in pre-clinical tumor models and in cancer patients.
