About Amit Kumar Singh Gautam
I am integrating my over 10 years of expertise in molecular biology, protein engineering, and assay development to create a transformative drug discovery platform addressing human diseases. Leading medium to high-throughput SPR/BLI-based screening platform for undruggable targets like transcription factors and GPCRs. Specializing in lentiviral vector engineering, genome-integrated reporter systems (AAVS1/CRISPR), and quantitative cell assays for target validation and therapeutic discovery. I utilized the power of display technology and a large library to solve the mystery of degradation initiation and leverage this information for targeted protein degradation. I have over 10 publications, invited talks, and provisional patents. Passionate about collaborative innovation, I strive to advance the field of drug discovery while driving impactful solutions for unmet medical needs.
Experience
Senior Scientist
May 2024 — Present · Cambridge, MA, US
Entrepreneur by nature, I thrive in a fast-paced environment. I am wearing multiple hats across design, molecular biology, biochemistry, operations, and lab safety. I partner closely with our AI team to turn designs into validated, decision-ready data.Leading AI-guided construct design and production: Co-develop peptide/miniprotein designs with ML partners to maximize expression, stability, and activity; translate in-silico predictions into buildable vectors and functional assays.Protein production and purification: used multiple platforms like in E.coli, HEKExpi, and cell-free system to express and purify high quality protein and miniptroteins using different scaffolds like hFC, GFP, etc and analytical tools. Manage CROs for bulk protein production and analytics. Leading Modular lentiviral libraries: Built Tet-inducible lentiviral libraries encoding designed peptides/miniproteins against undruggable targets; established expression/stability quantification in mammalian cells (flow cytometry, Cytation imaging, and luciferase reporters).Leading Genome-integrated reporters: Engineered AAVS1/CRISPR-based dual reporters (luciferase + fluorescence) for single-copy, or lentiviral-based stable readouts in human cells.Leading Binding assays: Developed a BLI screening workflow that quantifies target protein–peptide binding from crude lysate and purified components. Screened >500 binders for several difficult targets, including transcription factors and membrane protein targets (GPCRs -VLPs and Nanodisc)(15 20% hit rate calling). Determined full kinetics of hits and evaluated non-target and non-specific binding for the go/no-go decisions. Experience with high-throughput binding assays, including AlphaScreen/FP (384-well format). Experience with protein modeling tools like AlphaFold2, EMSFold, Rosseta, etc.Cross-functional delivery: Author SOPs, package clean data for program reviews, and manage CRO scope/timelines/QC to ensure on-time, high-quality deliverables.
