My journey into gene editing started during my PhD in 2013 where I generated my very first miRNA knockout fish model. Then, I joined the Wellcome Trust Centre for Human Genetics at the University of Oxford for a Postdoctoral Researcher postion and I focused my research on gene editing. The aim of the project was to develop new Cas9 variant that have the potential to reduce mosaicism with the hope to reduce the animal cost to generate a GA model. I have used a bench of Molecular and cellular techniques from protein production to NGS sequencing to achieve a complete characterization of the new variant both in animal model and cell lines.Technical skills-Over 10 years experience in genome editing in cells and animal models including fish and mouse (through embryo injection or ES cells)-More than 5 years working on in vivo gene editing, on mice embryos and blastocyst genotyping -Expertise in genome editing in mammalian cells including and mouse ES cells-2D and 3D differentation of iPS cells to model rare gentic diseases-FACS analysis and immunostaining to characterise cells or gene editing events-NGS based techniques, such as amplicon sequencing from library preparation to data analysis-Protein expression and purification-Extensive expertise in vector design including Traffic light reporter, Fluroscent based assay, mRNA and gRNA template and Cas9 variants
