Principal Scientist I - Cytometry and Tissue Imaging in Immuno-Oncology, Hematology, Translational Profiling- Member of a global team of translational/reverse translational immunologists, clinical scientists, chemical biologists, data scientists, hematologists, oncologists and facility operations to lead CyTOF single-cell analysis technology at NIBR in Cambridge- Operation, maintenance, training, and use of the CyTOF XT single-cell analysis platform including regular troubleshooting and communication with Tech Support, procurement of spare parts, software upgrade, daily performance, documentation of general guidelines for new CyTOF users- Development and optimization of new CyTOF 40+ antibody panels in collaboration with Diseases Area groups including panel design, reagents procurement, antibodies conjugation, staining procedures, and data analysis consultation with users regarding clinical studies and comparison between conventional or spectral flow cytometry validated panels - Performed new CyTOF 40+ antibody panels validation, basic data quality evaluation and deep single-cell analysis using licensed softwares (Flowjo, OMIQ, CellEngine)- Set up a quality control inventory of CyTOF reagents (antibody conjugates, viability and nuclei reagents) with documentation of procedures and resultant data electronically- Training of new CyTOF users for samples preparation (PBMCs, whole blood, dissociated tissues, cell lines) and acquisition on CyTOF XT- Trained on conventional flow cytometers (Agilent NovoCyte, BD LSRFortessa), Cytek Aurora spectral cytometer, and cell sorters instruments (BD FACSAria IIu, Sony MA900) within the NIBR Flow Core facility with fluidics start up, QC of the instruments for users (Lasers start up, stream set up, CS & T beads daily performance check of cytometer settings, RCP beads eight peaks for adjustment of PMT detector voltage dynamic range, manual drop delay, side stream trajectory, compensation with single color beads).
