Determining dynamics and transfer of the antibody repertoire upon vaccination in various donors (pregnant and newborn) via high-throughput pairing of the variable heavy and light chain B cell receptor (BCR) sequences from single cells via the emulsification of peripheral blood mononuclear cells (PBMC), followed by library preparation for NGS (Illumina NextSeq)-(BCR-Seq) to generate reference databases, and preparing serum and breastmilk samples via the pulldown of vaccine antigen-specific antibodies for mass spectrometry analysis - Immunoglobulin sequencing (Ig-Seq) under the framework of Maternal Immunization and Determinants of Infant Immunity (MADI) Consortium;• Guiding PhD students with various projects, including troubleshooting the isolation of antigen-specific B cell populations from primary samples for single cell RNA-Seq experiments for 10xGenomics, constructing cell lines using CRISPR-Cas9, Flp-in recombinase to create a mammalian antibody display system for antibody discovery; mentoring and instructing junior lab members including undergraduate students and research technicians (affinity purification using Protein G, Peptide M, His-Pur Co resins, ELISA, SDS-PAGE and agarose gel electrophoresis, buffer and media preparation);• Carrying the responsibilities of the laboratory safety officer and the cell sorter maintenance.
