Adipose Organoids- Generated adipose organoids with unilocular lipid morphology using SVF-derived cells. Established flow cytometry panels to characterize SVF populations, optimizing depot-specific differentiation- Performed bulk and single-nuclei RNA-seq to confirm all major adipose cell types and enhanced metabolic pathways- Developed organoid cross-sections, immunostaining panels to validate tissue-like architecture and and a lipolysis assay for functionality.Adipocyte Modeling – using ASC52telo- Engineered MSCs with adipocyte-specific transcription factors to achieve up to 99% differentiation into white, beige, and brown adipocytes- Established single-nuclei RNA-seq workflows, increasing nuclei yield by 10x. Applied siRNA and CRISPR-KO to induce thermogenically active beige adipocytes and assess transcriptional and functional changes- Developed a beige adipocyte ICC panel and partnered with imaging teams to integrate cells into high-throughput, imaging-based screening platforms.Hepatocyte Modeling – using iPSCs- Engineered iPSCs with hepatocyte-specific transcription factors and a custom small-molecule cocktail to increase mature hepatocyte yield to 75%- Converted 3D iHEP cultures into imaging-compatible monolayers- Conducted pooled optical CRISPR screening (POSH) for MASLD target discovery and established lipid accumulation assays with ML-driven phenotypic analysis- Evaluated Quantitative Phase Imaging (QPI) on 3D hepatocyte spheroids to identify cellular organelles and morphological features.
